[A new, in vitro technique for estimating the invasive potential of brain tumors].

چکیده
The authors previously reported a new, in vitro technique for determining the invasiveness of four rat neurogenic tumor cell lines. Now this method has been modified and applied to the analysis of the invasiveness of human brain tumors. Eighty-seven tumors, including 61 gliomas, six metastatic brain tumors, 10 meningiomas, four neurinomas, one melanoma, one hemangioblastoma, two craniopharyngiomas, one chemodec toma, and one pituitary adenoma, were studied. Since gliomas were of primary interest, their in vasiveness was studied more extensively. Fresh tumor specimens obtained during craniotomy were immediately minced in Dulbecco's modified Eagles's minimum essential medium containing 20% fetal calf serum to obtain fragments of 300-500 μm in size. These tumor fragments and precultured embryonic chick heart fragments were placed together on semisolid agar (0.35%) for 2 hours at 37°C. These fused tissues were then cultured in a gyratory shaker at 100 rpm for 7 days. From hematoxylin-eosin stained and immunostained serial sections, the degree of invasiveness was deter mined according to the “classification of i vasiveness.” Although there were discrepancies between the histologic findings and the degree of invasiveness in 22.5% of high-grade astrocytomas, con ventional histological diagnosis was well correlated with invasiveness in almost all low-grade astrocytomas. This in vitro technique appears to be a simple and useful means of evaluating the in vasive potential of brain tumors.

Estimating Tumor/Non-Tumor Uptake from Radiolabeled Monoclonal Antibodies using Scintigraphic Images and Dissecting the Animal Models
Introduction: Biodistribution study in animal models bearing tumors is one of the most important procedures in evaluation of fractional uptake of radiopharmaceuticals in the tumor and non-tumor organs. The aim of this study was to develop a new software-based method to determine activities that accumulate in the main organs as well as the tumor based on scintigraphy images, thereby obviating th...
متن کاملEvaluation of Nucleostemin Gene Expression as a New Molecular Marker in Breast Tumors
Background & Aims: Nucleostemin is one of the stem cell enriched proteins which encodes a novel nucleolar GTP-binding protein found at high levels in the adult and embryonic stem (ES) cells but not in terminally differentiated cells. It is also expressed in tumor cell lines as well as in the several types of human cancers. Due to the increasing rate of breast cancer in recent years, in the pres...

Gliomas account for almost 80% of primary malignant brain tumors in adults. Magnetic Resonance imaging (MRI) is still the gold standard for diagnosis of brain tumors and brain 99mTc-tetrofosmin Single Photon Emission Computed Tomography (99mTc-tetrofosmin-SPECT) has been established as a useful tool for their evaluation. Fluorine-18–2-fluoro-2-deoxy-d-glucose positron emi...
متن کاملAn overview of therapeutic approaches to brain tumor stem cells
Primary and secondary malignant central nervous system (CNS) tumors are devastating invasive tumors able to give rise to many kinds of differentiated tumor cells. Glioblastoma multiform (GBM), is the most malignant brain tumor, in which its growth and persistence depend on cancer stem cells with enhanced DNA damage repair program that also induces recurrence and resists current chemo- and radi...
متن کاملVascular Mimicry Expression in Invasive Ductal Carcinoma; A New Technique for Prospect of Aggressiveness
Background & Objective: In vascular (vasculogenic) mimicry (VM), tumoral cells mimic the endothelial cells and form the extracellular matrix-rich tubular networks. It has been proposed that VM is more extensive in aggressive tumors. This study was designed to investigate the rate of VM expression in the stromal cells of invasive ductal carcinoma (IDC) and to find its relationsh...
